It's certainly fairly vague but my ruling would be no, you cannot use Twinned Spell with Bigby's Hand. My main reasoning is that the casting of Bigby's Hand does not require a target. It just creates the hand. Thus it does not fulfil the "When you cast a spell that targets only one creature" portion of Twinned Spell.
